Could be a shadow process running it still, close/restart the host + virtual box?

You might try and different USB port as well to ensure its not tied up there

Host machine using it could tie it up if running simultaneously

Try removing any USB filters on the box (not sure if that's for vbox or the specific machines inside)

Also make sure if your loading a saved state to warm boot the box in case a shadow process is running
